Every time I leave "Manditraki Warrior" I am told that I have discovered 50% of the secrets in the level. What are the two "secrets"? I've combed the level (it's only three screens) and I don't see anything. Is this a glitch?

No glitch - have you tried "spin jumping" around?

Can you give me a hint how to enter the level and beat Manditraki Warrior? I cannot even think of a way to get in because I always get killed though I try everything.

Here's some hints to start you off...
When you first enter the level do some spin jumping. Next, go up on top of the pillars. Watch out for falling objects! Nothing in your arsenal will work, so don't waste your time trying to shoot.
