He probably meant STR instead of STR#.
yes they can. but the ID would be different. Use Pontus's EV Formula Calc

I use Shmelta-V to program missions for EVO. I switch them to EV, edit them, then switch them back to EVO. When I add Strings in shmelta, it makes them STR's instead of STR#'s. What I mean to say is does it make any difference for the EVO engine?

Much better! Now anyone can understand the question.

It should not matter to the EVO engine whether text is in 'STR ' or 'STR#' resources, as long as the correct ID numbers are used. If you will read the EVO Bible section "Appendix III - Patching STR# Resources" all (well, almost) will be explained.
