I got this mission I while back and, I was in no particular rush, so I did some other missions at the same time. Then I check my active missions and its gone. I have since completed many mission strings, and I am trying to get that on mission again. Is it possible? Any suggestions?

You probobly destroyed the ship she was on, depending on the leg of the mission you were on. If she's dead, you won't be able to get the mission again on your pilot.
Go to Knox, and see if you can get the mission again. If not... well... she's dead, I guess.

Is there some sort of plug I can make to revert it to the beginning? ie. convince the computer it never happened.

You could write a small mission that clears the mission bit that says you've failed the mission, but that would be cheating, according to some people.

Basically, you find out the mission bit that is set if you fail the mission, then make another mission that clears that bit. If you need help, look at the EVO Bible, and you can ask on the developer's board.

Not necessarily - all it means is that she and all her baggage etc. take up an amount of space that rounds up to one ton.
